Details for receptor: LXR-alpha

EndoNet ID: ENR01077

To link to the content of EndoNet use the EndoNet ID that is given on the detail pages in the format ENX0000, where X is a place holder for the type of the component (e. g. R for receptor or C for anatomical structure).
As URL for the linking append this ID to the detail page for this type of component.
For an hormone that would be:

http://endonet.bioinf.med.uni-goettingen.de/hormone/ENH00000

It is also possible to use the search of EndoNet to link to the right detail page. The URL should look like

http://endonet.bioinf.med.uni-goettingen.de/search/ENC00000
If the search pattern is unambigious the user is directed to the corresponding detail page.

General information

  • The adrenal gland expresses abundant quantities of both LXR-alpha and LXR-beta as well as the known LXR agonist 22(R)-OH cholesterol, an intermediate in the conversion of cholesterol to pregnenolone. [1]
  • Visceral organs specific expression, strong expression in liver, kidney and intestine followed by spleen and to a lesser extent the adrenals. [2]
  • Binds naturally occurring oxidized cholesterol derivatives such as 24(S),25-epoxycholesterol, 22(R)-, 24(S)-, and 27-hydroxycholesterol and synthetic nonsteroidal compounds such as T-0901317. [3]
  • Regulation of hepatic cholesterol homeostasis is mediated by the liver X receptors (LXR-alpha and LXR-beta). [1]

    • fat_cell

      Induced phenotypes

      • regulation of gene expression
        • Expression and ligand activation of LXRα has no significant effect on adipogenesis or lipid accumulation and does not modulate the adipogenic activity of PPARγ. The identification of novel LXR adipocyte target genes in cultured cells and in vivo points to an important role for LXR in the regulation of adipocyte gene expression and adipose tissue function distinct from PPARγ. [4]
      • regulation of lipid biosynthetic process
        • LXR activates the coordinate expression of major fatty acid biosynthetic genes (lipogenesis) and increased plasma triglyceride and phospholipid levels. [5]

        • Co-transfection of SREBP-1c downregulates APOA5 promoter activity. [6]
      • positive cholesterol
        • Liver X receptor (LXR) alpha and beta are the nuclear receptors responsible for regulation of cholesterol metabolism. In physiological conditions, high intracellular cholesterol levels cause increased synthesis of oxysterols, which activate LXR, thus triggering a transcriptional response for cholesterol secretion and catabolism. [7]

      Induced phenotypes

      • positive regulation of fatty acid biosynthetic process
        • LXR as a master lipogenic transcription factor, as it directly regulates both sterol regulatory element-binding protein 1c (SREBP-1c) and carbohydrate response element-binding protein (ChREBP) to enhance hepatic fatty acid synthesis. [8]
